An immunofluorescence assay for studying replication of adeno-satellite virus.

چکیده
The simian adeno-satellite virus apparently cannot multiply unless host cells are co-infected with a 'helper' adenovirus. Following simultaneous infection of green monkey kidney cells with simian adenovirus (SV 15) and its satellite, satellite antigen was first detected by immunofluorescence in the nucleus 10 to 12 hr after inoculation, while adenovirus antigen was first detected at 16 hr. A single cycle of growth for satellite virus was completed in about 24 to 48 hr. Inoculation of satellite-flee adenovirus from 10 to 15 hr before inoculation with satellite proved to be the most efficacious time for shortening the latent period of satellite. Satellite antigen could then be detected as early as 4 hr after inoculation. These results indicate that events in the adenovirus replication cycle must proceed for 10 to 12 hr before satellite can be synthesized. An infectivity titration of satellite based on immunofluorescence was developed. The percentage of fluorescent cells in standard monolayers was determined and the infectious units were calculated from the dilution infecting 1 ~ of cells. Most satellite preparations had titres of l0 s to 109 infectious units/ml, with one infectious unit equivalent to 30 to 100 particles.

Buthionine Sulfoximine Inhibits Cytopathic Effects and Apoptosis Induced by Infection with AIK-HDC Strain of Measles Virus
Measles virus (MV) is a highly contagious agent which causes a major health problem in developing countries. We studied the effect of buthionine sulfoximine (BSO) on the replication of an AIK-HDC strain of MV and its induced apoptosis in Vero cell lines. Methods: In this study, toxicity of BSO on Vero cells was investigated first, resulted in determination of sub-lethal or non-toxic concentrati...
متن کاملThe Effect of DNA-Dependent Protein Kinase on Adeno-Associated Virus Replication
BACKGROUND DNA-dependent protein kinase (DNA-PK) is a DNA repair enzyme and plays an important role in determining the molecular fate of the rAAV genome. However, the effect this cellular enzyme on rAAV DNA replication remains elusive. METHODOLOGY/PRINCIPAL FINDINGS In the present study, we characterized the roles of DNA-PK on recombinant adeno-associated virus DNA replication. Inhibition of ...
متن کاملIn vitro replication of adeno-associated virus DNA.
An in vitro assay for adeno-associated virus (AAV) DNA replication has been developed. The substrate is a plasmid containing the duplex form of AAV DNA in pBR322. The AAV insert is excised or rescued from the plasmid by extracts of uninfected cells. Replication was assayed by production of full-length excised AAV DNA resistant to Dpn I digestion. The following results were obtained. (i) Only ex...
متن کاملThe adeno-associated virus rep gene inhibits replication of an adeno-associated virus/simian virus 40 hybrid genome in cos-7 cells.
A hybrid adeno-associated virus (AAV)/simian virus 40 (SV40) genome is described. In this construct SV40 regulatory sequences, including the early promoter/enhancers and origin of DNA replication, were substituted for the AAV p5 promoter, which normally controls expression of the AAV rep gene. The hybrid genome was phenotypically indistinguishable from wild-type AAV in human cells in the presen...
متن کاملPomegranate peel extract inhibits internalization and replication of the influenza virus: An in vitro study
Objective: Influenza virus, which is associated with high level of morbidity and mortality, has been recently considered a public health concern; however, the methods of choice to control and treat it are limited. Our previous study showed anti-influenza virus activity of pomegranate peel extract (PPE). In this study, the mechanism through which PPE acts against influenza virus...
